| Abstract | Rhizophora mucronata L. mangrove leaves contain various secondary metabolites, especially flavonoids and phenolic compounds, which have the potential as natural antioxidants. This study is aimed to identify secondary metabolite compounds and determine the antioxidant activity of 70% ethanol extract and fractions of mangrove leaves (Rhizophora mucronata L.). The study was conducted through extraction using 70% ethanol, followed by fractionation with n-hexane, ethyl acetate, and water solvents. Identification of secondary metabolites was carried out using the Thin Layer Chromatography (TLC) method, while antioxidant activity was tested by the DPPH method using a UV-Vis spectrophotometer based on the IC₅₀ value. TLC results showed the presence of alkaloids (Rf 0.30), flavonoids (Rf 0.75), tannins (Rf 0.63–0.75), saponins (Rf 0.56–0.62) and steroids (Rf 0.30–0.50). The Rf value obtained was close to the standard reference and previous studies, thus confirming the presence of secondary metabolites in mangrove leaves. The results of the antioxidant test showed that the IC₅₀ value of the ethyl acetate fraction was 82.577 µg/mL, the water fraction was 89.435 µg/mL, the 70% ethanol extract was 94.623 µg/mL, and the n-hexane fraction was 102.308 µg/mL. The ethyl acetate fraction had the highest antioxidant activity, so fractionation affected the antioxidant activity of mangrove leaves (Rhizophora mucronata L.). | 1 |
