| Abstract | Melastoma malabathricum L. leaves contain flavonoid compounds with various pharmacological activities. The determination of total flavonoid content by UV-Visible spectrophotometry is influenced by several factors, including the pH of the solution. This study is aimed to evaluate the effect of pH variation on the total flavonoid content of 70% ethanol extract of Melastoma malabathricum L. leaves and to determine the optimum pH for analysis. This laboratory experimental study used the maceration method with 70% ethanol for extraction. Total flavonoid content was determined using the aluminum chloride (AlCl₃) colorimetric method with quercetin as the reference standard. The pH conditions tested were 2, 4, 6, 8, and 9.5. The analytical method was validated for linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ). Statistical analysis was performed using One-Way ANOVA followed by Tukey's HSD test. The results showed that pH variation significantly affected the total flavonoid content (p<0.05). The total flavonoid contents at pH 2, 4, 6, 8, and 9.5 were 26.230, 38.609, 43.516, 45.676, and 44.731 mg QE/g extract, respectively. The highest total flavonoid content was obtained at pH 8, indicating that this pH was the optimum condition for determining total flavonoid content using UV-Visible spectrophotometry.
