Abstract ABSTRACT Suciwulan, C. Pharmacological Evaluation of the Antihyperglycemic and Antioxidant Activity of Ethanol Extract from Salak Fruit Peel (Salacca zalacca) (Gaertn.) Voss in Induced Diabetic Rats. 2026, Bachelor of Pharmacy Study Program, Faculty of Pharmacy, Deli Husada Deli Tua Health Institute. (Supervised by apt. Anggun Syafitri, S.Farm., M.K.M., M.Farm.). Diabetes mellitus is a metabolic disease whose main factor is related to oxidative damage due to increased Reactive Oxygen Species (ROS). Oxidative stress occurs due to an imbalance between ROS and antioxidants in the body. Antioxidants play an important role in protecting cells from free radical damage, thus potentially preventing chronic diseases such as diabetes. One plant that has the potential as an antidiabetic is snake fruit peel because it contains flavonoid compounds. This study aimed to determine the antihyperglycemic and antioxidant activity of ethanol extract of snake fruit peel (Salacca zalacca (Gaertn.) Voss) in alloxan-induced rats by comparing its effectiveness to glibenclamide and determining the IC₅₀ value of ethanol extract of snake fruit peel. The study was conducted experimentally using 25 white male rats (Wistar strain) which were divided into five groups, namely negative control Na-CMC 1%, positive control glibenclamide 0.45 mg/kgBW, and group of ethanol extract of snake fruit peel doses of 100 mg/kgBW, 200 mg/kgBW, and 400 mg/kgBW. Antioxidant testing was carried out using the DPPH method using 96% ethanol as a solvent. The results of statistical analysis showed that the ethanol extract of snake fruit peel was able to reduce blood glucose levels significantly and had an effectiveness equivalent to glibenclamide. A dose of 400 mg/kgBW showed the most optimal antihyperglycemic effect. The antioxidant test results showed an IC₅₀ value of 16.96 ppm, which is considered a very strong antioxidant. The conclusion of this study is that the ethanol extract of snake fruit peel has the potential as an alternative antidiabetic therapy. The suggestion from this study is that the future researchers are expected to conduct toxicity testing on the ethanol extract of snake fruit peel.
