| Abstract | Sambung Nyawa (Gynura procumbens (Lour.) Merr) plant contains secondary metabolite compounds such as alkaloids, flavonoids, tannins, saponins, and steroids, which are capable of inhibiting antibacterial activity. This study aims to determine the antibacterial activity of mouthwash formulation containing Ethanol Extract of Sambung Nyawa Leaves against the growth of Escherichia coli bacteria at concentration variations of 10%, 20%, and 30%, as well as to evaluate the physical quality of the preparation. This study used an experimental method by testing the physical evaluation parameters of the mouthwash formulation, including organoleptic, pH, and viscosity tests. The results of the physical evaluation showed that the mouthwash formulations at concentrations of 10%, 20%, and 30% exhibited excellent physical stability, consistent green color, distinctive aroma, and met the safe requirements for oral mucosa. Based on the antibacterial activity test using the disc diffusion method, the sambung nyawa leaf extract mouthwash was able to inhibit the growth of Escherichia coli bacteria. The measurement results showed that the average diameter of the inhibition zone at 10% concentration was 8.68 mm (moderate category), 20% concentration was 9.20 mm (moderate category), and 30% concentration provided the largest inhibition zone reaching 10 mm (strong category). The Minimum Inhibitory Concentration (MIC) value achieved in this study was demonstrated at a concentration of 30% (showing clear tube results). | 1 |
