| Abstract | The asam gelugur leaf (Garcinia atroviridis) is a plant that contains secondary metabolites such as alkaloids, flavonoids, saponins, tannins, and steroids, which have potential antibacterial properties. This study aimed to determine the content of secondary metabolites and the antibacterial activity of ethanol extracts and leaf fractions of asam gelugur against Escherichia coli, as well as to identify the most effective concentration for inhibiting the growth of this bacterium. The study was conducted experimentally. The extraction of asam gelugur leaves using the maceration method with 96% ethanol as the solvent, followed by fractionation using n-hexane and ethyl acetate as solvents. Antibacterial activity was tested using the disk diffusion method at concentrations of 40%, 60%, and 80%, with amoxicillin serving as the positive control. The inhibition zone diameters were analyzed using one-way ANOVA, followed by Tukey’s post hoc test. The results showed that the crude extract contained alkaloids, flavonoids, saponins, tannins, and steroids. The n-hexane fraction contained steroids; the ethyl acetate fraction contained alkaloids, flavonoids, and tannins; while the residue fraction contained alkaloids, flavonoids, saponins, and tannins. The ethanol extract and all fractions exhibited antibacterial activity against Escherichia coli. An 80% concentration was the most effective, with the ethyl acetate fraction producing the largest inhibition zone diameter (20,1 mm). Based on these results, it can be concluded that the ethanol extract and fractions of asam gelugur leaves have potential as antibacterial agents against Escherichia coli. Further research is suggested to develop an ethyl acetate fraction of gelugur leaves into a pharmaceutical formulation, test it against other types of bacteria, and determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) to further investigate its antibacterial potential.
