Abstract Free radicals are one of the major causes of oxidative stress, which contributes to the development of various degenerative diseases. One approach to overcoming this problem is the utilization of natural antioxidants derived from plants, one of which is raru bark (Cotylelobium melanoxylon). This study aimed to determine the antioxidant activity of the ethyl acetate extract of raru bark (Cotylelobium melanoxylon) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. This study was conducted using an experimental research design. The extract was obtained through the maceration method using ethyl acetate as the extraction solvent, followed by characterization, phytochemical screening, and antioxidant activity testing using the DPPH method. Absorbance measurements were performed using a UV-Visible spectrophotometer to determine the IC₅₀ value, and the analytical method was validated through linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ) tests. The results showed that the ethyl acetate extract of raru bark contained flavonoids, alkaloids, tannins, saponins, and steroids, which potentially contribute to its antioxidant activity. The extract exhibited an IC₅₀ value of 74.5812 ppm, indicating strong antioxidant activity. The validation results demonstrated that the DPPH method fulfilled the required validation parameters and was suitable for antioxidant activity analysis. In conclusion, the ethyl acetate extract of raru bark (Cotylelobium melanoxylon) has the potential to serve as a natural antioxidant source and may be further developed as a raw material for pharmaceutical products and functional foods. These results suggest that further studies employing other antioxidant assay methods, including ABTS, FRAP, and CUPRAC, as well as investigations of other parts of Cotylelobium melanoxylon, are warranted to provide a more comprehensive evaluation of its antioxidant potential. 1
