| Abstract | Seven golden candlestick (Cassia alata L.) is a traditional medicinal plant empirically used to treat various skin diseases such as ringworm, scabies, and fungal infections. This efficacy is thought to be related to the flavonoid content of its leaves. This study aimed to identify the presence of flavonoid compounds and determine the total flavonoid content in the ethanol extract of Cassia alata L. leaves using UV-Vis spectrophotometry. This was a descriptive quantitative study with a laboratory approach. Leaf samples were obtained from Bulan-Bulan Village, Batu Bara Regency, extracted by maceration using 96% ethanol, followed by qualitative testing and determination of total flavonoid content using quercetin as the reference standard. The qualitative test results showed that the ethanol extract of Cassia alata L. leaves was positive for flavonoids, alkaloids, saponins, and tannins. UV-Vis spectrophotometric measurement yielded a maximum wavelength of 432 nm with a calibration curve equation of y = 0.07698x + 0.02593 and a correlation coefficient (r) of 0.9932. The total flavonoid content of the ethanol extract was 5.6387% (quercetin equivalent, %w/w). Method validation showed good accuracy with an average recovery of 96.97%, good precision with a %RSD of 0.0023%, and LOD and LOQ values of 0.7402 mg/L and 2.2431 mg/L, respectively. Based on these results, the UV-Vis spectrophotometry method was declared valid for determining the total flavonoid content in the ethanol extract of Cassia alata L. leaves. Further research is recommended to investigate the pharmacological activities and optimize the extraction conditions of Cassia alata L. leaves. | 1 |
