Abstract Diabetes mellitus is a metabolic disorder characterized by elevated blood glucose levels due to impaired insulin secretion or action. Bandotan leaves (Ageratum conyzoides L.) contain secondary metabolites that may have antidiabetic effects, making them a potential natural ingredient for helping to lower blood glucose levels. This study aimed to determine the effectiveness of ethyl acetate extract from bandotan leaves in lowering blood glucose levels in male white rats with alloxan-induced diabetes and to identify the most effective dose. The study was conducted experimentally using male white rats divided into a negative control group (0.5% Na-CMC), a positive control group (0.45 mg/kg body weight glibenclamide), and treatment groups receiving bandotan leaf ethyl acetate extract at doses of 100, 200, and 300 mg/kg body weight. Blood glucose levels were measured periodically over 24 days. The data were analyzed using a paired t-test and one-way ANOVA, followed by Tukey's test. The results showed that administration of the ethyl acetate extract of bandotan leaves at doses of 100, 200, and 300 mg/kg body weight reduced blood glucose levels by 73.45%, 74.41%, and 76.54%, respectively, while the glibenclamide positive control reduced blood glucose levels by 78.38%. Statistical analysis revealed significant differences among the treatment groups (p<0.05). Based on the study results, it can be concluded that the ethyl acetate extract of bandotan leaves is effective in lowering blood glucose levels in male white rats with alloxan-induced diabetes, with 300 mg/kg body weight being the most effective dose. Further research is recommended to develop the ethyl acetate extract of bandotan leaves in various dosage forms and at different doses, conduct toxicity evaluations, and perform histopathological examinations to assess pancreatic ẞ-cell regeneration.
