| Abstract | Phyllanthus emblica L. (Malacca fruit) is rich in secondary metabolites, lites, including flavonoids, alkaloids, tannins, saponins, and steroids, which are known to possess antibacterial activity. This study aimed to evaluate the antibacterial activity of the ethanolic extract of Phyllanthus emblica L. fruit against Escherichia coli and multidrug-resistant Escherichia coli (MDREC). Antibacterial activity was assessed using the broth microdilution method in a 96- well microplate to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). In addition, bacterial DNA and protein leakage assays were performed to investigate the antibacterial mechanism. The results showed that the ethanolic extract of Phyllanthus emblica fruit exhibited MIC values of 4,000 µg/ml against Escherichia coli and 8,000 µg/ml against multidrug- resistant Escherichia coli (MDREC), while the MBC value for both bacterial strains was 16,000 µg/ml. The DNA and protein leakage assays demonstrated increased absorbance with increasing extract concentrations, indicating bacterial cell membrane damage that resulted in the leakage of intracellular components. These findings suggest that the ethanolic extract of Phyllanthus emblica L. fruit possesses antibacterial activity against Escherichia coli and multidrug-resistant Escherichia coli (MDREC) by disrupting the bacterial cell membrane. | 1 |
