| Abstract | Mikania micrantha Kunth (sembung rambat) and Clidemia hirta L. (harendong) leaves contain secondary metabolites such as flavonoids, alkaloids, tannins, saponins, steroid, and terpenoids that possess potential antibacterial activity. A combination of these two extracts is expected to provide superior antibacterial activity against Staphylococcus epidermidis, a bacterium associated with acne. This study is aimed to formulate a serum preparation combining the ethanolic extracts of Mikania micrantha kuth and Clidemia hirta leaves, determine the physical characteristics of the preparation, and assess its antibacterial activity against Staphylococcus epidermidis. This experimental study involved several stages: preparation and characterization of crude plant material (simplicia), phytochemical screening, extraction via maceration using 96% ethanol, formulation of the serum using three concentration variations F1 (5%:10%), F2 (10%:5%), F3 (10%:10%), evaluation of physical quality (organoleptic properties, homogeneity, pH, and spreadability), and antibacterial activity testing against Staphylococcus epidermidis using the diffusion method. The obtained data were statistically analyzed to identify the formula with the best physical characteristics and the most effective antibacterial activity. The results of this study are expected to provide scientific information regarding the potential of combining ethanolic extracts of Mikania micrantha kuth and Clidemia hirta leaves as active ingredients in a serum preparation that meets physical quality standards, is safe for use, and effectively inhibits the growth of Staphylococcus epidermidis.
