Abstract Peningkatan resistensi bakteri terhadap antibiotik sintetik mendorong eksplorasi bahan alam sebagai alternatif antibakteri, salah satunya daun bakau kurap (Rhizophora mucronata L.) yang dilaporkan mengandung flavonoid, alkaloid, tanin, saponin, serta steroid/terpenoid. Nilai Rf ekstrak (0,75) mendekati kuersetin (0,71), mengonfirmasi keberadaan flavonoid. The increasing resistance of bacteria to synthetic antibiotics has prompted the exploration of natural products as alternative antibacterial agents, including the leaves of Rhizophora mucronata L., which have been reported to contain flavonoids, alkaloids, tannins, saponins, and steroids/terpenoids. This experimental study aimed to evaluate the the antibacterial activity of ethanol extract nol extract of R. mucronata leaves against Staphylococcus aureus and Staphylococcus epidermidis, and to identify flavonoid compounds using Thin Layer Chromatography (TLC). The herbal drug was characterized according to the Indonesian Herbal Pharmacopoeia and extracted by maceration with 96% ethanol. The extract was subsequently subjected to phytochemical screening, flavonoid identification by TLC, and antibacterial testing using the disk diffusion method at concentrations of 10%, 20%, and 30%. Data were analyzed using One-Way ANOVA followed by Tukey's HSD test. Results showed that the herbal drug met characterization standards, and the extract tested positive for flavonoids, alkaloids, tannins, saponins, and steroids/terpenoids. The Rf value of the extract (0.75) was close to that of quercetin (0.71), confirming the presence of flavonoids. The extract inhibited the growth of both bacteria, with inhibition zones increasing with concentration; the highest inhibition zones at 30% were 12.95 mm (S. aureus) and 13.13 mm (S. epidermidis), classified as strong. ANOVA revealed significant differences among treatments (p<0.05), and Tukey's HSD test confirmed 30% as the most effective concentration. In conclusion, the ethanol extract of R. mucronata leaves exhibits antibacterial activity against S. aureus and S. epidermidis, with 30% as the optimal concentration, supported by the presence of flavonoid compounds identified by TLC. 1
