| Abstract | This study aimed to determine the effectiveness of a mouthwash based on nanosimplicia of black cumin seeds (Nigella sativa L.) in inhibiting the growth of Streptococcus mutans bacteria. Streptococcus mutans is one of the main causes of dental caries, so an alternative natural ingredient with antibacterial activity is needed. Black cumin seeds are known to contain secondary metabolite compounds such as alkaloids, flavonoids, saponins, tannins, terpenoids, and anthraquinones which have potential as antibacterial agents. This study used a laboratory experimental method. Black cumin seed simplicia was characterized, then phytochemical screening was carried out and it was processed into nanosimplicia using the sonication method. The nanosimplicia was then formulated into a mouthwash preparation with concentration variations of 5%, 7.5%, and 10%. The preparations were evaluated including organoleptic test, homogeneity, pH, viscosity, particle size, polydispersity index, and zeta potential. The antibacterial activity against Streptococcus mutans was tested using the disc diffusion method and the results were statistically analyzed using the One Way ANOVA test followed by the Tukey test. The results showed that the black cumin seed simplicia met the characterization requirements based on the Indonesian Herbal Pharmacopoeia and contained alkaloid, flavonoid, saponin, tannin, terpenoid, and anthraquinone compounds. All nanosimplicia formulas had particle sizes in the nanoparticle range (134.20–284.10 nm), while the formula with a concentration of 10% showed the smallest particle size (134.20 ± 1.66 nm) and the highest zeta potential (32.70 ± 0.35 mV). The results of statistical analysis showed a significant difference between formulas (p<0.05). The higher the concentration of black cumin seed nanosimplicia used, the better the characteristics of the nanoparticles and the greater the potential for antibacterial activity against Streptococcus mutans. | 1 |
