Abstract Furunculosis is an acute skin infection of the hair follicles, commonly caused by the pathogenic bacteria Staphylococcus aureus. The problem of antimicrobial resistance to standard therapies such as mupirocin has prompted the exploration of alternative antibacterial agents based on natural ingredients. This study aimed to determine the antibacterial activity of ethanol extracts of soursop (Annona muricata L.) and cherry (Muntingia calabura L.) leaves, both singly and in combination, against Staphylococcus aureus bacteria and to compare their effectiveness with a commercially available preparation (Mupirocin 2%). The research method used was a laboratory experiment. Soursop and cherry leaf samples were extracted by maceration using 96% ethanol. Characterization of the medicinal plants and phytochemical screening were performed prior to microbiological activity testing. Antibacterial activity testing was performed using the Kirby-Bauer disc diffusion method with varying concentrations of the single and combined extracts (5%, 10%, and 15%), a positive control of 2% mupirocin, and a negative control of 70% ethanol. Data from the inhibition zone diameter measurements were analyzed descriptively and statistically using a one-way ANOVA followed by a. Phytochemical screening results showed that both extracts contained post-hoc test secondary metabolites including alkaloids, flavonoids, tannins, saponins, and steroids. The antibacterial activity test for the single soursop leaf extract yielded inhibition zone diameters of 5.55 mm (5%), 8.63 mm (10%), and 13.32 mm (15%). The single cherry leaf extract produced inhibition zones of 6.41 mm (5%), 7.44 mm (10%), and 8.35 mm (15%). The combination of the two extracts produced a significant synergistic effect, with inhibition zone diameters reaching 13.58 mm at a 5%:5% concentration, 14.70 mm at a 10%:10% concentration, and 16.81 mm at a 15%:15% concentration. The 15%:15% combination concentration showed strong antibacterial activity, almost comparable to the positive control of 2% mupirocin (17.11 mm), while the negative control showed no inhibition zone (0 mm). The conclusion of this study indicates that the combination of ethanol extracts of soursop leaves and cherry leaves has more optimal antibacterial activity than either of the two forms alone and has the potential to be developed as an alternative therapy for furunculosis. 1
