Abstract Free radicals are molecules containing unpaired electrons that can induce oxidative stress and contribute to the development of various degenerative diseases. One potential source of natural antioxidants is avocado seed (Persea americana Mill.), which contains secondary metabolites such as flavonoids, phenolics, tannins, and saponins. This study aimed to evaluate the antioxidant activity of the 96% ethanol extract of avocado seeds and to determine the fraction with the strongest antioxidant activity using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method. This experimental study involved the preparation of simplicia, extraction by maceration using 96% ethanol, fractionation with n-hexane, ethyl acetate, and water, followed by antioxidant activity testing using a UV-Visible spectrophotometer at a wavelength of 516 nm. Antioxidant activity was expressed as the IC₅₀ value obtained from the linear regression between sample concentration and percentage inhibition. The results showed that the ethanol extract of avocado seeds exhibited antioxidant activity with an IC₅₀ value of 72.72 ppm, which was categorized as strong. The n-hexane fraction showed an IC₅₀ value of 93.43 ppm (strong), while the ethyl acetate fraction exhibited an IC₅₀ value of 43.12 ppm, indicating very strong antioxidant activity. The water fraction demonstrated lower antioxidant activity value of 235,43 ppm. As a positive control, vitamin C exhibited an IC₅₀ value of 11.2 ppm, which was categorized as very strong. Based on these findings, it can be concluded that the ethyl acetate fraction possessed the highest antioxidant activity because it had the lowest IC₅₀ value among all tested fractions. This result suggested to the next researchers to encouraged to employ different extraction methods and further develop the ethanol extract of avocado seeds into pharmaceutical formulations or health products with added value and potential for commercialization. 1
