Abstract Excessive exposure to ultraviolet (UV) radiation can trigger the formation of free radicals, leading to cellular skin damage, uneven pigmentation and premature aging. Durian (Durio zibethinus L.) seeds are a food waste product containing potential secondary metabolites such as flavonoids, phenolics, alkaloids, saponins, and steroids/triterpenoids that act as natural antioxidants. This study is aimed to formulate an ethanolic extract of durian seeds into a stable skin-lightening lotion and to evaluate its antioxidant activity using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. Durian seed samples were extracted via maceration using 96% ethanol, yielding 10.173% extract. The extract was then formulated into lotions at varying concentrations: F0 (base 0%), F1 (2.5%), F2 (5%), and F3 (10%). The resulting lotions underwent physical stability evaluations (organoleptic properties, homogeneity, pH, spreadability, and viscosity), safety testing (irritation) on 15 volunteers and skin-lightening efficacy testing over 14 days (days 0, 1, 7, 10, and 14) using a digital skin analyzer. Antioxidant activity was measured via UV-Vis spectrophotometry at the maximum wavelength (517 nm), using Vitamin C as a reference standard. Physical evaluation results indicated that all lotion formulas were stable, homogeneous, and met the quality parameters for topical preparations. The formulations exhibited a pH range of 4.5–6.5 (safe for skin physiology) and optimal spreadability of 5–7 cm and caused no irritation (redness, itching, or swelling) on the panelists' skin. Skin analyzer-based efficacy testing revealed significant improvements in skin tone (brightness), hydration (water content) and softness following regular application, particularly with the F3 (10%) formula. Based on linear regression analysis, free radical scavenging tests demonstrated that the ethanolic extract of durian seeds possesses active antioxidant capacity, which correlates with its potential as a skin-brightening agent through the mitigation of oxidative stress. 1
