| Abstract | Dayak onion leaves (Eleutherine palmifolia (L.) Merr.) are one of the medicinal plants native to Central Kalimantan and have been traditionally used by the Dayak community as herbal medicine. The leaves are known to contain various secondary metabolites, including flavonoids, alkaloids, tannins, and saponins, which are believed to possess antibacterial activity. This study is aimed to evaluate the antibacterial activity of the ethanol extract and the n-hexane, ethyl acetate, and aqueous fractions of Dayak onion leaves against Staphylococcus aureus. The extraction was carried out using the maceration method with 96% ethanol, then continued with liquid–liquid fractionation using n-hexane, ethyl acetate, and water. Antibacterial activity was evaluated using the disc diffusion method at concentrations of 20%, 40%, and 60%. The results showed that the ethanol extract and all fractions of Dayak onion leaves exhibited inhibitory activity against Staphylococcus aureus. Among the tested samples, the ethyl acetate fraction at a concentration of 60% produced the largest inhibition zone. These findings indicate that Dayak onion leaves have the potential to be developed as a natural antibacterial agent against Staphylococcus aureus. Keywords: Eleutherine Palmifolia (L.)
