| Abstract | Exposure to free radicals and ultraviolet (UV) radiation is one of the major factors contributing to skin damage and premature aging. Mangosteen peel (Garcinia mangostana L.) and wild betel leaf (Piper aduncum L.) possess potential antioxidant and photoprotective properties. This study aimed to formulate a gel serum containing a combination of ethanolic extracts of mangosteen peel and wild betel leaf, evaluate its physical characteristics and stability, and determine its antioxidant activity and Sun Protection Factor (SPF) in vitro. The plant materials were extracted by maceration using 96% ethanol, yielding extraction recoveries of 18.64% for mangosteen peel and 11.14% for wild betel leaf. Phytochemical screening revealed that the ethanolic extract of mangosteen peel contained alkaloids, flavonoids, saponins, tannins, and triterpenoids, whereas the ethanolic extract of wild betel leaf contained alkaloids, flavonoids, saponins, tannins, and steroids. The gel serum was formulated into four formulations consisting of F1 (3%:1%), F2 (4%:1.5%), F3 (5%:2%), and F4 (5%:2%) combinations of the extracts. The results demonstrated that all formulations met the required physical quality parameters, including acceptable organoleptic properties, homogeneity, pH compatible with skin pH, spreadability, adhesiveness, drying time, viscosity, and stability under temperature stability testing. Antioxidant activity evaluated using the DPPH method showed IC₅₀ values of 76.07 µg/mL (F1), 45.21 µg/mL (F2), 40.04 µg/mL (F3), and 37.62 µg/mL (F4). The SPF values were 1.58 for F1 (ineffective), 19.03 for F2, 23.34 for F3, and 28.85 for F4, with F2, F3, and F4 classified as providing ultra protection. Formula F4 exhibited the strongest antioxidant activity, as indicated by the lowest IC₅₀ value, and the highest SPF value among all formulations. | 1 |
