| Abstract | Acne is an inflammatory skin disease primarily associated with the growth of Cutibacterium acnes. Excessive use of antibiotics may lead to bacterial resistance; therefore, natural antibacterial agents are needed as alternative treatments. Pumpkin peel (Cucurbita moschata D.) and green betel leaf (Piper betle L.) contain various secondary metabolites, including alkaloids, flavonoids, tannins, saponins, steroids, and glycosides, which are known to possess antibacterial properties. This study aimed to evaluate the antibacterial activity of ethanol extracts of pumpkin peel, green betel leaf, and their combinations against Cutibacterium acnes, the bacterium associated with acne development. This experimental study employed a maceration extraction method using 96% ethanol as the solvent. Antibacterial activity was assessed using the disc diffusion method at concentrations of 20%, 40%, 60%, 80%, and 100%, as well as extract combinations at ratios of 25:75, 50:50, and 75:25. Clindamycin was used as a positive control, while 70% ethanol served as a negative control. Phytochemical screening revealed the presence of alkaloids, flavonoids, tannins, saponins, steroids, and glycosides in both extracts. Microscopic identification showed Gram-positive bacteria with short rod-shaped to pleomorphic morphology, consistent with the characteristics of Cutibacterium acnes. The ethanol extract of pumpkin peel produced inhibition zones ranging from 9.25 to 13.16 mm with an MIC value of 0.5474, whereas the ethanol extract of green betel leaf produced inhibition zones ranging from 17.83 to 21.41 mm with an MIC value of 0.0072. The best combination was observed at a ratio of 25:75, producing an inhibition zone of 21.25 mm, although it remained lower than clindamycin (22.58 mm). It can be concluded that green betel leaf ethanol extract exhibited the strongest antibacterial activity against Cutibacterium acnes, while the combination of both extracts did not demonstrate a synergistic effect. | 1 |
