Abstract Rhizophora mucronata L., commonly known as bakau kurap, is a mangrove plant that contains various secondary metabolites with potential antibacterial properties. This study aimed to evaluate the antibacterial activity of the ethanolic extract of Rhizophora mucronata leaves against Escherichia coli and Bacillus subtilis, determine the most effective extract concentration in inhibiting bacterial growth, measure the inhibition zone diameter, and identify the secondary metabolites using Thin Layer Chromatography (TLC). This experimental study was conducted by preparing simplicia from bakau kurap leaves, followed by extraction using the maceration method with 96% ethanol as the extraction solvent. The antibacterial activity was evaluated using the disc diffusion method at several extract concentrations, with an antibiotic as the positive control and dimethyl sulfoxide (DMSO) as the negative control. Identification of secondary metabolites was performed using Thin Layer Chromatography (TLC) based on the retention factor (Rf) values and the color of chromatographic spots observed under ultraviolet light at 254 nm and 366 nm. The results demonstrated that the ethanolic extract of Rhizophora mucronata leaves exhibited antibacterial activity against Escherichia coli and Bacillus subtilis, as indicated by the formation of inhibition zones at each extract concentration. TLC analysis also revealed the presence of several groups of secondary metabolites that were presumed to contribute to the antibacterial activity of the extract. In conclusion, the ethanolic extract of Rhizophora mucronata leaves possesses antibacterial activity against Escherichia coli and Bacillus subtilis and contains secondary metabolites that can be identified using Thin Layer Chromatography (TLC), indicating its potential as a natural source of antibacterial agents. 1
