| Abstract | Mangrove plants, particularly Rhizophora mucronata, are recognized as a potential source of bioactive compounds, especially flavonoids, which possess antioxidant and various pharmacological activities. However, information regarding the flavonoid profile and total flavonoid content of the ethanolic extract of bakau kurap leaves is still limited. Therefore, qualitative identification and quantitative determination of flavonoids are necessary to provide scientific data supporting the utilization of this plant as a natural medicinal resource. This study aimed to qualitatively identify the flavonoid profile using the Thin Layer Chromatography (TLC) method and quantitatively determine the total flavonoid content of the ethanolic extract of bakau kurap leaves (Rhizophora mucronata) using UV-Visible spectrophotometry. The study began with plant identification, preparation of simplicia, characterization of the simplicia, phytochemical screening, and extraction using the maceration method with 70% ethanol as the solvent. Flavonoid identification was carried out using Thin Layer Chromatography (TLC), while the determination of total flavonoid content was performed using the aluminum chloride (AlCl₃) colorimetric method with quercetin as the reference standard and analyzed using a UV-Visible spectrophotometer. The results showed that the yield of the ethanolic extract of bakau kurap leaves was 12.39%. TLC analysis showed that the sample had an Rf value of 0.75, while the quercetin standard had an Rf value of 0.96, indicating the presence of flavonoid compounds in the extract. The quercetin calibration curve produced the regression equation y = 0.055371x + 0.005733 with a coefficient of determination (R²) of 0.9996, indicating excellent linearity. The total flavonoid content of the ethanolic extract of bakau kurap leaves was 13.02 mg QE/g extract. Method validation demonstrated an accuracy of 100.14%, a precision with a limit of detection (LOD) of 12.984, and a limit of quantification (LOQ) of 43.28, indicating that the analytical method fulfilled the validation requirements. In conclusion, the ethanolic extract of bakau kurap leaves (Rhizophora mucronata) contains flavonoid compounds with a total flavonoid content of 13.02 mg QE/g extract. The TLC method successfully identified the presence of flavonoids, while the UV-Visible spectrophotometric method using aluminum chloride (AlCl₃) reagent proved to be valid and reliable for the determination of total flavonoid content in the ethanolic extract of bakau kurap leaves. It is recommended that future studies identify flavonoid compounds more specifically using advanced analytical techniques such as High-Performance Liquid Chromatography (HPLC) or Liquid Chromatography–Mass Spectrometry (LC-MS), as well as evaluate the biological activities of the extract to support the further development and utilization of bakau kurap (Rhizophora mucronata) leaves as a natural resource in the pharmaceutical field.
