| Abstract | Mahogany leaves (Swietenia macrophylla) contain secondary metabolites such as flavonoids, alkaloids, saponins, tannins, and steroids, that possess potential antibacterial properties. This study aimed to evaluate the physical characteristics of anti-acne patch formulations containing an ethanolic extract of mahogany leaves and to assess their antibacterial activity against Cutibacterium acnes at extract concentrations of 7.5%, 15%, and 22.5%. This was a laboratory-based experimental study patch evaluations included organoleptic properties, thickness, weight uniformity, folding endurance, pH, and irritation potential. The results demonstrated that all formulations met the established physical evaluation criteria. Antibacterial testing showed that the anti-acne patches containing mahogany leaf ethanolic extract were capable of inhibiting the growth of Cutibacterium acnes. Formulation F3 containing 22.5% extract exhibited the highest antibacterial activity, with a mean inhibition zone diameter of 14.60 ± 0.05 mm, followed by F2 (15%) at 10.54 ± 0.25 mm and F1 (7.5%) at 8.38 ± 0.16 mm. The study concludes that increasing the concentration of mahogany leaf ethanolic extract enhances antibacterial activity against Cutibacterium acnes, with F3 being the most effective formulation. Further research is recommended to evaluate the stability of the formulations under various storage conditions and to explore a wider range of extract concentrations.
