| Abstract | This study aims to identify secondary metabolite compounds through Thin-Layer Chromatography (TLC) profiling and evaluate the in vitro antibacterial activity of loop-root mangrove (Rhizophora mucronata L.) leaf ethanolic extract against Staphylococcus aureus and Escherichia coli. The mangrove leaf sample was extracted via maceration by using 96% ethanol. Phytochemical screening and TLC analysis confirmed the presence of secondary metabolites, including alkaloids, flavonoids, and tannins. Antibacterial activity testing was conducted using the well diffusion method at extract concentrations of 50%, 75%, and 100%, with Amoxicillin as the positive control and DMSO as the negative control. The results demonstrated that the ethanolic extract of loop-root mangrove leaves had significant effectiveness in inhibiting the growth of both test bacteria with a strong inhibition category. The average inhibition zone diameters formed against Staphylococcus aureus were 13,75 mm (50%), 15,26 mm (75%), and 16,40 mm (100%), respectively. Meanwhile, the average inhibition zone diameters against Escherichia coli were 12,26 mm (50%), 13,50 mm (75%), and 14,11 mm (100%), respectively. Based on these findings, it can be concluded that the ethanolic extract of loop-root mangrove (Rhizophora Mucronata L.) leaves has great potential to be developed as a natural antibacterial agent. | 1 |
