Abstract Infections caused by Staphylococcus aureus bacteria are still a health problem because they can cause various diseases and the increasing resistance to antibiotics. Therefore, alternative antibacterial agents from natural sources are needed. One potential natural source is raru stem bark (Cotylelobium melanoxylon) because it contains secondary metabolites such as flavonoids, tannins, and saponins, which are known to have antibacterial activity. This study aimed to determine the antibacterial activity of raru stem bark extract and to compare the effectiveness of hot extraction (reflux) and cold extraction (maceration) methods against Staphylococcus aureus bacteria. This experimental study used 96% ethanol extract of raru stem bark. The extracts were obtained using reflux and maceration methods. Phytochemical screening was carried out to identify the active compounds contained in the extract. Antibacterial activity was tested using the disc diffusion method by measuring the diameter of the inhibition zone formed against the growth of Staphylococcus aureus. The results showed that raru stem bark extract contained flavonoids, tannins, and saponins. Both reflux and maceration extracts showed antibacterial activity against Staphylococcus aureus. The highest antibacterial activity was obtained at a concentration of 60%, where the reflux extract produced an inhibition zone of 19.3 mm, while the maceration extract produced an inhibition zone of 18.5 mm. Both extracts were categorized as having strong antibacterial activity, but the reflux extract showed higher antibacterial effectiveness compared to the maceration extract. Based on these results, it can be concluded that raru stem bark (Cotylelobium melanoxylon) has potential as an antibacterial agent against Staphylococcus aureus. The reflux method produced better antibacterial activity than the maceration. Further research is recommended to investigate the antibacterial activity of other parts of the raru plant, develop raru stem bark extract into a more practical and standardized dosage form, and use a wider range of concentrations with the reflux method to determine the maximum inhibition activity.
